Structural-functional organization of FXR A) The schematic representation of the FXR domains structure B) FXR forms a heterodimer with retinoid X receptor (RXR) that binds to the FXR-response element (FXRE), a DNA sequence in the promoter of its target genes. In the absence of ligands, the FXR heterodimer is associated in complex with corepressor, leading to transcriptional repression. Following BAs binding to the LBD of FXR, the heterodimer undergoes conformational changes, leading to the recruitment of co-activators to replace corepressors, which results in the transactivation of target genes expression.
