Silver staining of SDS-PAGE gels showing structural changes to human plasma FN treated with MPO/H2O2/Cl in the presence of increasing concentrations of SCN. Purified human plasma FN (0.2 μM in 0.1 M phosphate buffer, pH 7.4) was either left untreated (MPO only control) or treated with MPO/H2O2/Cl and the indicated concentrations of SCN for 2 h at 37 °C. Samples were electrophoresed using 3–8% Tris-acetate SDS-PAGE gels under: A) non-reducing, or B) reducing conditions. Gels were then fixed, and visualized with silver staining and referenced against HiMark™ pre-stained high molecular mass standards. Black arrow = dimer/higher aggregates; white arrow = monomer bands.
