The RQ mutation increases PKG kinase activity in intact cells. A, 293T cells were cotransfected with expression vectors for Myc-tagged VASP and either WT or R177Q PKG1α. At 24 h posttransfection, some cells were treated with 100 μm 8-pCPT–cGMP for one hour. Cell lysates were analyzed by SDS-PAGE/immunoblotting using antibodies recognizing Myc-epitope (upper) or PKG1 (lower).
