Figure 1.

Inhibitory effects of shikonin on THP-1-CM-induced migration and invasion of A549 and H1299 cells. (A) Chemical structure of shikonin. (B) Effect of shikonin on the viability of lung adenocarcinoma cells. A549 and H1299 cells were treated with serial doses of shikonin in normal medium for 24 h, and the viability of cells was detected by CCK-8 assay. (C) A549 and H1299 cells were incubated with THP-1-CM and/or shikonin for 24 h, and the viability of cells was examined by CCK-8 assay. (D) Wound healing assays. A549 and H1299 cells were scratched and incubated with THP-1-CM and/or shikonin, and images were captured at 0 and 24 h after wounding. (E) Invasion assays were performed by Transwells covered with Matrigel. After treatment with THP-1-CM and/or shikonin for 24 h, the cell suspensions were subjected to Transwell assays. Images (magnification ×200) are representative of three independent experiments. Data are shown as the mean ± SD of three independent experiments. *P<0.05, **P<0.01, compared with the THP-1-CM-treated group. THP-1-CM, THP-1 cell conditioned medium.
