Effects of shikonin on the migration and epithelial-mesenchymal transition (EMT) of A549 and H1299 cells induced by IL-6 and TNF-α. (A) Transwell chamber migration assay. After treatment with shikonin and IL-6 (50 ng/ml) or TNF-α (20 ng/ml) for 24 h, A549 and H1299 cell suspensions were subjected to a Transwell chamber migration assay. (B) Western blot and (C) real-time PCR assays were used to detect the protein and mRNA levels of E-cadherin, N-cadherin, vimentin and Snail in A549 and H1299 cells after treatment with shikonin and IL-6 (50 ng/ml) for 24 h. (D) Western blot and (E) real-time PCR assays were used to detect the protein and mRNA levels of E-cadherin, N-cadherin, vimentin and Snail in A549 and H1299 cells after treatment with shikonin and TNF-α (20 ng/ml) for 24 h. β-actin was used as an internal control.
