Validation of methanol-based peroxidase inactivation for mIHC. (A) CD68 mIHC visualization (left panel) shows specific and strong staining in human tonsil tissue. Following CD68 visualization, chromogen was stripped and HRP was inactivated using hydrogen peroxidase (0.6%) diluted in 100% methanol. AEC was reapplied and no chromogenic signal was detected (B), confirming efficient and complete stripping of CD68 signal. (C) Then, CD3 antibody was detected with secondary and AEC chromogen, showing distinct, specific, and strong signal. Scale bars=500μm (low magnification) and 50μm (high magnification).
