DU325 induces differentiation of HL-60 promyelocytes. As a proof of cellular differentiation, the expression of AML blast and leukemia stem cell marker CD33 decreased (A). Matured myeloid cell marker CD11b elevated on the cell surface detected by flow cytometry (B). Due to maturation, the myeloperoxidase (MPO) activity of HL-60 cells increased upon treatment after 48 h (C). Cells were assayed as described in the Materials and Methods Section 4.12.4. for CD33 and CD11b immunostaining, and 4.11. for the MPO activity. Data are shown as arithmetic mean values ± standard deviation from triplicate experiments. Statistical significance was calculated in relation to untreated cells and set to ** p < 0.01, *** p < 0.001.
