Figure 1.

Secoemestrin C inhibits Con A-stimulated splenocyte proliferation and elevates SOCS proteins mRNA expression. (A) Chemical structure of secoemestrin C (SC) isolated from Aspergillus nidulans. (B, C) Splenocytes from C57BL/6J mice were stained with CFSE and stimulated with Con A (2 μg·mL-1) in the presence of secoemestrin C (SC) at the indicated concentration, and the cells were cultured for 72 h. Splenocyte proliferation was detected by flow cytometry. A representative histogram (B) and summary bar graph (C) are shown. (D) Splenocytes isolated from C57BL/6J mice were stimulated with Con A (2 μg·mL-1) in the presence of secoemestrin C (128 nM), and the cells were cultured for 24 h. The mRNA levels of SOCS1, SOCS2 and SOCS3 were measured by real-time PCR. Data are shown as means ± SEM (n=5 per group). *P<0.05, **P<0.01 and ***P<0.001 vs. the control group; ##P<0.01 and ###P<0.001 vs. the Con A-treated group.
