Inhibitory effects of the CDOTs on human platelet aggregation. Washed platelets (3.6 × 108 cells/mL) were preincubated with PBS (control) or the synthesized CDOTs (25-120 μM) and subsequently treated with (A,B) 1 μg/mL collagen, (C) 1 μM U46619, and (D) 0.01 U/mL thrombin to induce platelet aggregation. The IC50 and maximal inhibitory concentrations are shown in B. The inhibitory profiles (AD) are representative examples of four similar experiments. The delayed lag phase of platelet aggregation noticed in CDOT-pretreated platelets stimulated by either U46619 (C) or thrombin (D).
