Long-term adipogenic differentiation mediated fat accumulation into the porcine intramuscular preadipocyte (PIP) in vitro culture. The PIP cells (2.5 × 104/cm2) were kept in vitro culture for 20 days with differentiation medium, and cells were harvested at different time points to estimate the degree of cytosolic fat accumulation by using a triglyceride (TG) assay kit (A). Data shown are the mean ± SD of three independent experiments performed in triplicates. Different letters (a, b, c, and d) labeled upon the time points indicate significant differences (p < 0.05), while the same letter indicates no significant difference between the contrast pairs. Oil red O staining of adipocytes demonstrated the fat droplets red color while the nucleus takes a blue color when observed under microscope (B).
