CLEM imaging of AnxA6-GFP-expressing LHCN myotubes during the formation of the cap subdomain. (A) LHCN myotubes were transfected with pA6-GFP plasmid and membrane damage was performed at the edge of the myotubes, which were then rapidly fixed. (B) Recruitment of AnxA6-GFP was checked by fluorescence microscopy as described in the legend of Figure 6. (C) Myotubes were then immunostained for AnxA6 with a secondary antibody coupled with a gold nanoparticle and observed in TEM. Red and white arrows, irradiated area.
