Entry is not the determining factor to whether productive replication is initiated at 41 °C. (a) Schematic detailing experimental protocol. Primary CK cells seeded in 6 well plates were inoculated in triplicate with 105 PFU of IBV (MOI ~0.01). Cells were incubated for 1 h at either 37 °C (blue) or 41 °C (red), after which the inoculum was removed and the cells washed to remove unbound virus. Cells were incubated at either 37 °C or 41 °C for 23 h. The quantity of infectious viral progeny produced was assessed through titration in CK cells. Graph (b) represents data from H120, (c) D388(QX), (d) Beau-R and (e) M41-CK infection. Each point represents the mean from three independent experiments (n = 3) with error bars indicating SEM. Statistical differences were assessed using a one-way ANOVA with a Tukey test for multiple comparisons and are highlighted by ** (p < 0.005) and **** (p < 0.0001).
