The general reaction mechanism of aromatic prenylation catalyzed by class I terpene cyclases is shown. If the indole were flipped, then N-prenylation would occur. The position of indole in the active site pocket is based on the three-dimensional model in Fig. 6, which in turn is based on the experimentally determined position of imidazole in Fig. 5b.
