(A) Representative bright-field images of PLum-AD and PLum-AI prostatospheres in different conditions embedded in MatrigelTM at day 12 after plating. Scale bar = 50 μm. Images were visualized by Axiovert inverted microscope and analyzed by Carl Zeiss Zen 2012 image software. Prostatospheres formed in each well were counted at day 12 for both cell lines. (B) Treatment with SB203580 (10 μM) significantly decreased sphere forming units (SFUs) while IL-6 (10 ng/mL) increased it in both cell lines. (C) SB203580 significantly reduced the average diameter of spheres while IL-6 increased it in PLum-AD and PLum AI. Data represents the average diameter (μm) of 90 prostatospheres in each condition from three independent experiments. All data is reported as mean ± SEM (*P < 0.05, **P < 0.01 and ***P<0.001; every treatment condition was compared to its control, Student’s t-test).
