(B) Mapping of the regions in hnRNP K that interact with the FMDV IRES. Cell lysates prepared from HEK293T cells overexpressing FLAG-tagged wild-type hnRNP K or its truncated mutants were purified with an anti-FLAG G1 affinity resin and subjected to a biotin-RNA pulldown assay using the full-length FMDV IRES. Both the crude lysate (input) and proteins bound to the streptavidin beads (labeled RNA) were resolved by SDS-PAGE and detected by Western blotting using an anti-HA antibody. Unlabeled RNA was used as a negative control.
