Immunogenicity of Ad5-GUCY2C-S1 and Ad5.F35-GUCY2C-S1. (A–D) BALB/c mice (n=4–7 mice/group) were immunized intramuscularly with control or 1010 vp of Ad5-GUCY2C-S1 or Ad5.F35-GUCY2C-S1 and serum and splenocytes were collected 14 days later. GUCY2C-specific CD8+ T-cell responses were quantified by interferon gamma (IFN-γ) ELISpot (A) and antibodies were quantified by ELISA (B). (C) GUCY2C-specific T-cell avidity measurements were analyzed by ELISpot using non-linear regression (log(agonist) versus normalized response) with comparisons made using the extra sum-of-squares F test.
