Schematic of PLGA-encapsulated GSI Nanoparticles in the liver. GSI NPs accumulate in the liver, where they release GSI over time. Inhibition of γ-secretase prevents translocation of the Notch intracellular domain (NICD) into the nucleus, blocking target gene transcription. Downstream effects of Notch signaling inhibition include reduced macrophage (MΦ)-mediated inflammation, hepatic stellate cell (HSC)-induced fibrosis, and hepatocyte (H) glucose production.
