T4 TXTL synthesis as a function of PEG8000 concentration. Standard TXTL reactions were carried out with optimum settings: 5 mM magnesium, 60 mM potassium, 1 nM genome. Maximum TXTL T4 synthesis occurs around 3% PEG. The phage yields varies by orders of magnitude, as observed previously for the other phages ΦX174 and T7 (10).
