Effects of miR-483-3p on hFOB1.19 cell proliferation and osteogenic differentiation. (A) miR-483-3p overexpression was generated in hFOB1.19 cells by transfection with miR-483-3p, as confirmed by RT-qPCR. hFOB1.19 cells were transfected with miR-483-3p and examined for (B) cell viability by CCK-8 assay; (C) DNA synthesis capacity by EdU assay, and (D) cell apoptosis by flow cytometric assay. (E) The protein levels of Ki-67, cleaved-caspase-3, caspase-3, WNT1, β-catenin, cyclin D1, RANKL and OPG were examined by western blot analysis, and (F) the mRNA expression of osteogenesis markers, including ALP, RUNX2, OCN and OPN was determined by RT-qPCR. (G) The mineralization of hFOB1.19 cells was determined by Alizarin Red staining. *P<0.05, **P<0.01.
