Tube formation assay with HDAC4 overexpression. (A) HUVECs were transfected with GFP or HDAC4. After 24 h cells were seeded on matrigel. (B + C) HMECs were transfected with GFP or HDAC4. After 24 h cells were treated with TGFβ (10 ng/ml) or H2O2 (30 μM) for 24 h and seeded on matrigel. Representative images after 4 h of tube formation are shown. Numbers of nodes were counted and total tube length was measured with an image angiogenesis analyzer (Carpentier, 2012) in ImageJ and summarized in the statistics (n > 4).
