miR-4654 promotes the formation of autophagosomes in HSFBs following resveratrol treatment. HSFBs were infected with adenoviruses expressing GFP-LC3 fusion protein to obtain a stable GFP-LC3 cell line. Different treatments/transfections were applied in the stable cell line, including resveratrol treatment alone, resveratrol + miR-4654 mimic transfection, resveratrol + miR-4654 mimic + Rheb OE transfection, resveratrol + miR-4654 inhibitor transfection + Rheb KD, resveratrol + miR-4654 inhibitor transfection and the control group. After 72 h, DAPI was used to stain cells and the cells were observed at a magnification of ×200 using confocal microscopy. Scale bar=20 µm. The GFP-LC3 fluorescence intensity was normalized against the control group and determined using Image J software.
