Defining resident human leukocyte antigen (HLA)-DRINTCD11c+ dendritic cells (DCs) in spleen and lymph nodes (LNs) as well as migratory HLA-DRHIGHCD11c+ DCs in blood of adeno-associated virus serotype 9 (AAV9)/hucytokines human immune system (HIS) mice upon recombinant adenovirus serotype 5 (rAd5) vaccination. (A) Experimental scheme to generate and vaccinate HIS mice. Eighteen weeks (126 days) post-hematopoietic stem cell (HSC) infusion, HIS mice were vaccinated with AdPfCS and Adp24. One day later, CD11c+ DCs were isolated from blood, spleen, and LNs of rAd5-immunized and naive HIS mice, and the level of HLA-DR expression by CD11c+ DCs was determined. (B) Two subsets, HLA-DRINT and HLA-DRHIGH, were identified, and their frequency (%) was analyzed by pooled blood or tissue samples from 3 to 5 HIS mice per group, which were given injections containing human HSCs from the same donor. (C) Floating bars that express minimum, maximum, and mean values were overlaid by scatter graphs. The vaccination experiments were repeated twice and results represent 1 experiment (n = 3). Statistical analyses were performed using analysis of variance followed by Dunnett’s test, and differences were considered if P < .05.
