Transcription-based snapshot reporters.

The Cal-Light and FLARE Ca2+ biosensors create an “AND gate” design by combining the optogenetically controlled AsLOV2 domain with a Ca2+ switch controlled split-TEV protease, which leads to the cleavage of the tTa-VP16 transcriptional activator only in the presence of high Ca2+ and blue light347,348. The cleavage of the transcriptional activator will then turn on the expression of a reporter gene such as GFP expression.
