Effect of dCas9-sgRNAs on TCF4 expression levels. (A) Schematic with quantitative PCR (qPCR) primers for TCF4 mRNA and intronic RNA transcripts. (B) The qPCR results showing that dCas9-(CAG)n sgRNA reduces TCF4 intron two RNA levels and has no effects on TCF4 mRNA. Levels of TCF4 mRNA were assessed by measuring expression levels of constitutive exon 18 found in all isoforms. Results are shown as the mean ± SD, n = 2 independent experiments. P value was obtained by t-test analysis of NTC compared with dCas9-(CAG)n sgRNA.
