Changes in the signaling network of colorectal cancer (CRC) liver metastases upon decorin delivery. Liver-specific transfection with the DCN gene inhibited several RTKs with a concomitant decrease in the activity of the MAPK/ERK and Akt/mTOR/p70S6K pathways. Increased AMPK (possibly via LKB1) and p38/MSK/CREB signaling resulted in elevated phospho-p53 levels; stabilization of this key tumor suppressor may result in a cell cycle blockade. Inhibition of STATs and c-Jun proteins also contributes to the antitumor action of decorin. Color code: red: decreased; green: increased; orange: no change; grey: not examined.
