G100 inflames TME of A20 tumors. (A) A20 tumors (n = 4 mice/group) treated with G100 (10 μg/ml GLA, 2%SE) for 1 week (3 injections, every other day) were collected for gene expression analysis by Nanostring. Multiple immune response pathways, including type 1 interferon signaling, DC function, T cell, and NK cell functions, were all activated. (B) Tumor infiltrating lymphocytes (TIL) were collected for Flow cytometry analysis and stained with antibodies for CD3, CD4, CD8, Foxp3, and NKp46. G100 treatment induced significant increase in T cells and NK cells and a decrease in Treg among CD4 T cells. SE treatment was no different from PBS control in any of the markers.
