Panel III: sensing Nt.BbvCI and HindIII by the sensing module, switches on the fluorescence of FAM and Cy5. Panel IV: multiplexed sensing of all three endonucleases: Nt.BbvCI, EcoRI, and HindIII, switches on the fluorescence of FAM, ROX, and Cy5. In all experiments, the concentrations of Nt.BbvCI, EcoRI, and HindIII corresponded to 0.2, 0.2, and 2 U μL–1, respectively.
