Uptake mechanism in U87MG cells as a function of RGD format on the nanoparticle surface. Nanoparticles were prepared from PLGA-Rho in single-emulsifier experiments (i.e. 100% F127-OH or 100% F127-RGDx); their uptake was measured by flow cytometry on live cells after pre-treatment for 30 min and co-incubation with different agents and nanoparticles for 1 h at 37 °C, and expressed in relation to the control experiment performed without inhibitors (i.e. PBS-treated cells). Please note that all cells showed evidence of internalization (see Supplementary Fig. 6SI.B), they just differed in the amount of uptake. RGDc = cyclic RGD peptide, specifically targeting αvβ3 heterodimers. RGDl = linear RGD peptide, binding to both αvβ3 and α5β1 heterodimers. Statistical analysis (One-way ANOVA), n = 3.
