TAM promotes miRNA cleavage but not miRNA binding. (A) In vitro AGO1 cleavage assay shows that target RNA is cleaved when the TAM is present. The target RNAs were incubated for 1 or 2 h, where two cleavage products were present in the target with TAM on the X-ray film (as indicated). The asterisk indicates the background bands present in both control and experiment groups. The control represents the experiment at time 0. (B) In vivo AGO1 binding assay shows no difference between target RNAs with TAM and without TAM. RENILLA and ACTIN were used as the control. miRNA156 levels were measured in all the samples.
