Crocetin reduced the nuclear translocation of NF-kB in the livers of DENV-infected mice. BALB/c mice were infected with DENV-2 (4 × 105 FFU/mL) and then treated with crocetin (50 mg/kg) dissolved in 2% DMSO. DENV-infected 2% DMSO-treated and mock-infected 2% DMSO-treated groups of mice were used as positive and negative controls, respectively. Protein samples were prepared from liver tissues, and cellular fractionation was conducted. The cytosolic and nuclear fractions of the proteins were subjected to Western blot analysis, and the results were normalized to that of GAPDH (cytosolic housekeeping gene) and Lamin B1 (nuclear housekeeping gene). Densitometry analysis of blots was conducted using ImageJ software normalized to the individual housekeeping genes. (A) Western blot analysis of NF-kB p65 and p50 from the cytosolic fraction.
