At high ktet,max values, kcat is decoupled from the rate of the tethered reaction and may disguise large differences in the tethered phosphorylation rate. (B) Fitting curves from Figs. 2C (WT) and 2E (R-3K) show unequal increase in ktet (2.1× and 9.0×, respectively) upon increasing Ceff from 100 to 1,000 µM.
