SP biosynthesis is directly coupled to Gnp1-mediated serine uptake. The SPT pool at the cortical ER is highly regulated by serine uptake and the TORC1/Ypk1/Orm1/2 signaling network. The nuclear envelope localized SPT pool is responsible for constant SP biosynthesis. This model adds serine uptake as another factor to previously described mechanisms regulating SP homeostasis by the signaling cascade of Slm1/2-TORC2-Ypk1/2 and the Orm proteins [7–9].
