The difference between a true variant and an artifact from a UMI perspective. (A) A true variant is present on the DNA fragment so when the UMI tag 1 is added, it tags the fragment and the mutation as well. After amplification, all the fragments tagged with the UMI tag 1 carry the same mutation. (B) An artifact is not present on the DNA fragment but rather appears at the steps that follow the UMI introduction. That is why not all fragments with the same UMI tag 2 carry the same artifact.
