Polyanhydride nanoparticles induce CD8+ T cell memory with a low antigen dose. C57BL/6 female mice were immunized subcutaneously with formulations consisting of 75 μg soluble Ova plus 500 μg of 20:80 CPTEG:CPH polyanhydride nanoparticles encapsulating 25 μg Ova (NP) (n=12); 100 μg soluble Ova alone (sOva) (n=11); or PBS control (n=12). Mice were challenged subcutaneously in the flank with E.G7 Ova-expressing tumor cells 42 days post-immunization and (A) tumor volume of individual mice was tracked. (B) Survival was evaluated 30 days post-challenge. Significance from PBS control was determined using a Log rank (Mantel-Cox) test with a Bonferroni correction for multiple (2) comparisons. Significance from naïve control group is indicated in the treatment group legend as follows *p ≤ 0.025. Median survival of each group is also indicated parenthetically.
