IL23 is required for optimal induction of antimicrobial pathways upon chronic NOD2 stimulation of human macrophages. MDMs were transfected with scrambled, IL23R, or IL12Rβ2 siRNA, and then treated with 100 μg/mL muramyl dipeptide (MDP). (A) After 48 hours of treatment, S Typhimurium–GFP uptake was assessed (n = 8, similar results in an additional 4 donors). Significance was between the IL23R siRNA- and scrambled siRNA-transfected MDMs for the corresponding treatment condition. (B) Fold phospho-PDK1 at 15 minutes (n = 4). (C–G) ROS production and induction of the indicated proteins at 48 hours (n = 4) (mean fluorescence intensity [MFI]). (H) Fold cathepsin D activity at 48 hours (n = 12 from 2 independent experiments). Means + SEM. ∗P < .05, ∗∗∗P < .001, †P < 1 × 10-4, and ††P < 1 × 10-5. Scr, scrambled.
