Screening of PRV open reading frames (ORFs) that modulate the cGAS–STING-induced transactivation of the IFN-β promoter. PK15 cells were transfected with IFN-β-Luc (200 ng), pCMV-RL (2 ng) mixed with pcDNA4-HA-cGAS (50 ng), pcDNA3-Flag-STING (20 ng), pcDNA3-Flag (200 ng), or pcDNA3-Flag expressing PRV ORFs (200 ng). The total amount of DNA was normalized using the pcDNA3-Flag and pcDNA4-HA empty vectors (E.V) in the negative control. The cells were collected 30 h post-transfection and then analysed for luciferase activity. The fold change in relative luciferase activity is calculated as the luciferase activity induced by cGAS–STING with or without each PRV ORF divided by that induced in the pcDNA3-Flag control group.
