Recruitment of Pol II to viral DNA is defective in the presence of SP-2509. MRC-5 cells were infected at an MOI of 10 PFU/cell with strain KOS in the presence or absence of SP-2509 (16 μM). EdC was added at 4 hpi and cells were fixed at 6 hpi. Viral genomes were covalently attached to a fluorophore (vDNA) and probed with antibodies specific for the indicated proteins. For Pol II, two different colocalization phenotypes were observed. The top row (with SP-2509) represents the colocalization of Pol II with replicated viral DNA, which occurred in 33 out of 51 counted cells (65%), while the bottom row represents the lack of colocalization of replicated viral DNA with Pol II, which occurred in 18 out of 51 counted cells (35%).
