Concept of the E. coli surface display based on post‐translational SC‐ST coupling. The membrane anchor Lpp‐OmpA is genetically fused to a SC domain, while the passenger is fused to the smaller ST. Subsequent to expression from individual plasmids, the fusion proteins covalently couple through a specific SC‐ST interaction. The complex is anchored in the E. coli outer membrane by Lpp‐OmpA.
