(A) The reaction of PARP-1 autoPARylation. NAM = nicotinamide; (B) AutoPARylation of recombinant PARP-1 induced by a-DNA (positive control from supplier). When Veliparib, an enzymatic inhibitor of PARP-1 (indicated with In), was added to the reaction mixture no PARylation was observed (negative control). The Western blot was performed with anti poly/mono ADP-ribose Ab and a secondary Ab conjugated to horseradish peroxidase. A membrane replica showing the protein markers (right) is also reported.
