High throughput system for applying mechanical forces to cultured cells. (A) The system applies mechanical strain to cells cultured in a 96 well plate format. A platen with 576 pistons is moved by a linear motor to displace a flexible membrane in six 96-well plates with flexible culture surfaces. Bar = 5 cm. (B) The culture plate consists of two plates that connect to hold a thin silicone membrane that serves as a culture surface. Thicker silicone membranes serve to seal each well and prevent leakage between the wells. Teflon pistons mounted on the platen are displaced into the membrane to apply mechanical strain to the cells cultured in each well.
