JTMT mice exhibit improved blood perfusion and angiogenesis in diabetic HLI. T2D was induced by HFD plus STZ in JTMT and littermate WT mice. The blood perfusion was monitored before and at 0, 3, 7, 14, 21, and 28 days after HLI using a PeriCam PSI (A) and quantified by Image J (B). Ischemic hind limb muscle tissues were then collected at day 28 after HLI. Transverse sections of gastrocnemius (GS) muscle (C) and soleus (SS) muscle (D) were stained for CD31 to enumerate CD31+-stained cell number as a proxy of capillary density. Capillary density was expressed as CD31+ capillaries per high-power field (HPF) (E), and DAPI was used to recognize nuclei.
