Comparison of the cost and time requirements for the plasmid and RCA systems. The cost calculations in the table are based on the following kits: Promega PureYield™ Plasmid Miniprep System (catalog A1222), ZymoPURE™ II Plasmid Midiprep Kit (catalog D4201), and Evomics SuperPhi RCA Premix Kit with Random Primers (catalog number PM100). In constructing the cost estimate, it was assumed that ten colonies were selected for screening, and one to four of those colonies were then midiprepped and sequenced.
