Mechanism of SSB and DSB induction following formation of ICLs. Depurination followed by slower elimination of the 3′ phosphate would introduce a SSB. Alternatively, hydrolysis of the AP site by APE1 in tissue culture would introduce a break in the DNA. Repetition of either pathway at the remaining intact alkylated nucleotide would form a DSB.
