PLD2 inhibitor promotes SAHA-induced suppression of angiogenesis. a Inhibitory effects of SAHA and PLD2 inhibitor on angiogenesis in MDA-MB-231 and MDA-MB435 CAM-implanted tumors. The cells were loaded (1.5 × 106 cells/CAM) onto CAMs and SAHA and/or PLD2 inhibitors were administered at the time of implantation. Five days after implantation, the CAM was resected and imaged under the microscope. Tumor vasculature was analyzed and the number of vessels determined. The data shown represent the mean ± SEM of at least six chick embryo CAMs. b The CAM of a 10-day-old chick embryo was exposed to vehicle or VEGF. PLD2 inhibitor and SAHA were treated on top of the CAMs by means of filter disks. After a 3-day incubation period, the CAM tissue directly beneath each filter disk was resected from the control and the drug-treated embryo CAMs. Digital images of CAM sections exposed to filters were captured. The data represent the mean ± SEM of at least six chick embryo CAMs. c MDA-MB231 cells were treated with SAHA and/or PLD2 inhibitor for 24 h, and the lysates were analyzed by western blot.
