One-pot combinatorial synthesis of short-chain aminolipids via double-reductive amination. (A) Double-reductive amination of aldehydes with amines in dichloromethane (DCM) in the presence of sodium triacetoxyborohydride (STAB) as a reducing agent. (B) Briefly, amines (Am) are mixed with excess aldehyde (Ald) and stirred at RT for 3 h followed by the addition of STAB. The reaction continues under stirring at RT for 3–5 days. The obtained aminolipids are then extracted using DCM/brine and then purified by column chromatography (silica, DCM/MeOH 98:2 → 90:10).
