Figure 3. — Validation of time-resolved miRNA expression patterns by TaqMan assays for independent donors. To confirm the validity of the time-resolved miRNA expression patterns of the microarray analyses (donors 1 and 2), the time-course experiments were reproduced with cells of four independent donors (donors 3, 4, 5 and 6). Time-course RNA samples were collected at 0, 2, 4, 8, 12 and 24 h after activation. As control, RNA samples was collected from non-activated cells after 24 h. Time-resolved miRNA expression patterns were analyzed by TaqMan assays. Donor 2 served as a reference of the microarray time-courses. A paired t-test was performed, comparing the time points of minimum and maximum expression that were determined based on the median results of the donors 3–6, assuming a normal distribution of the data.
