Mir-146b-5p Suppressed NF-κB Activity in Lung Cancer Cells

(A) The expression levels of miR-146b-5p in NF-κB reporter A549 cells were measured using qRT-PCR. The expression of miR-146b-5p was normalized to RNU6B expression. qRT-PCR data were presented as the mean ± SD (Student’s t test: ∗∗∗p < 0.001). (B, C) A549-κB cells were transiently transfected with miR-Ctl or miR-146b-5p mimics in the absence or presence of IL-1β. After 48 h of transfection, the activity of the NF-κB reporter was assessed using a dual-luciferase reporter assay system (B), and the nuclear NF-κB subunits (p50 and p65) were detected using western blot (C). The relative luciferase activities are presented as means ± SD from three independent experiments (Student’s t test: ∗∗p < 0.01, ∗∗∗p < 0.001). PBS, phosphate-buffered saline. (D) Relative expression levels of IL6 (left panel) and IL8 (right panel) mRNAs were determined after transfection of miR-Ctl or miR-146b-5p mimics in the PC9/gef, PE2988, or PE3479cells using qRT-PCR. qRT-PCR data were presented as the mean ± SD (Student’s t test: ∗p < 0.05, ∗∗∗p < 0.001).
