Effect of SPINK3 on Sodium-dependent depolarization induced by EGTA. CatSper opening was indirectly assessed by membrane potential (Em) recordings, and following the magnitude of the EGTA-induced depolarization. Capacitated (CAP) or non-capacitated (NC) sperm were loaded with 1 μM DISC₃(5) during Em recordings. (A) Representative traces are shown for all conditions assayed: CAP (green), NC (black), CAP + 13 μM SPINK3 (yellow); NC + 1 mM 8Br-cAMP (red); CAP + 13 μM SPINK3 + 1 mM 8Br-cAMP (blue).
