Parasites that lack AAP3.2 are unable to develop in mice. Late-stationary-phase L. donovani promastigotes of the WT and Δap3D6, Δap3D10, and Δap3G3-17 mutants were injected intravenously (i.v.) into 8-week-old female BALB/c mice (1 × 10⁸ cells per injection; eight mice per mutant). On day 21 postinfection, mice were sacrificed, and liver DNA was extracted using the proteinase K method (26). Analyses were carried out as described in Materials and Methods.
