Simplified protocol for single-cell transcriptomics or genomics, from the sample collection to sequencing. Briefly, the tissue sample is dissociated to obtain a single-cell suspension. An optional enrichment/sorting step could be required for rare cell types (dashed arrow). cDNA libraries are then prepared, barcoded to identify individual cells and individual transcripts and sequenced. If the gonadal cellular composition is more important, then 10× Chromium technologies if often chosen. If sequencing depth is more important, Smart-seq2 is typically preferred.
